The synthesis of collagen begins with the transcription of specific genes into messenger RNA (mRNA), which is then translated into procollagen molecules. These molecules undergo post-translational modifications, including hydroxylation and glycosylation, within the cell. Procollagen is then secreted from the cell, where it is cleaved into tropocollagen units. These tropocollagen molecules self-assemble into fibrils, which further aggregate to form mature collagen fibers. This hierarchical assembly is essential for the strength and integrity of connective tissues like skin, tendons, cartilage, and bone.

Collagen deposition is tightly regulated throughout life. During development, it forms the scaffolding for organs and tissues. In response to injury, cells like fibroblasts increase collagen production to repair damaged areas.
