Inflammasomes are multi-protein complexes that play a crucial role in the innate immune system's response to various stressors, including pathogens, damage-associated molecular patterns (DAMPs), and other danger signals. These complexes are responsible for the activation of inflammatory responses, particularly through the activation of caspase-1, an enzyme that processes pro-inflammatory cytokines such as interleukin-1β (IL-1β) and interleukin-18 (IL-18).

The term "inflammasome" was coined in 2002, and since then, several types of inflammasomes have been identified. The most well-studied inflammasomes include the NLRP3, NLRC4, AIM2, and Pyrin inflammasomes. Each inflammasome is composed of a sensor protein, which detects specific danger signals, an adaptor protein called ASC (apoptosis-associated speck-like protein containing a CARD), and the enzyme pro-caspase-1. Upon activation, these components assemble into a large complex, leading to the autocatalytic activation of caspase-1.

The activation of inflammasomes is a tightly regulated process that involves two signals. The first signal, often provided by pathogen-associated molecular patterns (PAMPs) or DAMPs, primes the cell by upregulating the expression of inflammasome components and pro-inflammatory cytokines. The second signal triggers the assembly of the inflammasome complex, leading to the activation of caspase-1 and the subsequent release of active IL-1β and IL-18.

Inflammasomes are involved in a wide range of physiological and pathological processes, including infection, autoinflammatory diseases, and metabolic disorders. Dysregulation of inflammasome activity has been implicated in various diseases, such as gout, type 2 diabetes, and Alzheimer's disease. Understanding the molecular mechanisms underlying inflammasome activation and regulation is therefore of great interest for the development of new therapeutic strategies.
