Immunoprecipitációval (IP) is a laboratory technique used to isolate a specific protein from a complex mixture, such as a cell lysate or serum. The process relies on the highly specific binding of an antibody to its target antigen.

The fundamental principle involves incubating a solution containing the protein of interest with an antibody that is known to bind to that specific protein. This antibody-antigen complex then needs to be separated from the rest of the solution. This is typically achieved by immobilizing the antibody onto a solid support, such as agarose beads or magnetic beads. These beads, coated with the antibody, are added to the protein mixture. The antibody binds to its target protein, forming a complex that is now attached to the beads.

Following the binding step, the beads are separated from the solution, usually by centrifugation or magnetic separation. The beads, along with the bound antibody and its target protein, are washed thoroughly to remove any non-specifically bound proteins. Finally, the bound protein is eluted from the antibody by disrupting the antibody-antigen interaction, often using a low pH buffer or a chaotropic agent. The eluted protein can then be further analyzed using techniques like Western blotting, mass spectrometry, or enzyme activity assays to confirm its identity and study its properties. Immunoprecipitation is a powerful tool in molecular biology and biochemistry for protein purification, detection, and characterization.
