The hypothalamuspituitary axis, often referred to as the HPA axis, is a central neuroendocrine system in the vertebrate body that regulates a wide range of physiological functions. It comprises the hypothalamus, a small region in the brain, and the pituitary gland, a pea-sized gland located at the base of the brain. These two structures are intricately linked and communicate through a complex feedback loop.

The hypothalamus produces releasing and inhibiting hormones that control the secretion of hormones from the anterior pituitary gland. In response, the anterior pituitary releases its own tropic hormones, which then act on various peripheral endocrine glands, such as the adrenal glands, thyroid gland, and gonads. The posterior pituitary, on the other hand, stores and releases hormones produced by the hypothalamus, namely antidiuretic hormone (ADH) and oxytocin.

This axis plays a crucial role in maintaining homeostasis. It is involved in regulating stress responses, growth and development, metabolism, reproduction, body temperature, and fluid balance. For instance, during stress, the hypothalamus releases corticotropin-releasing hormone (CRH), which stimulates the pituitary to release adrenocorticotropic hormone (ACTH). ACTH then signals the adrenal glands to produce cortisol, a stress hormone. The elevated levels of cortisol then feedback to inhibit the hypothalamus and pituitary, shutting down further CRH and ACTH release. Disruptions in the HPA axis can lead to various endocrine disorders and health problems.
