Hematoxylin is a natural dye commonly used in histology and microscopy. It is derived from the heartwood of logwood trees, primarily Haematoxylum campechianum, which are native to Central America. The active coloring agent in hematoxylin is hematoxylin, which is oxidized to hematein during the staining process.

Hematoxylin is typically used in combination with eosin in a technique called hematoxylin and eosin (H&E) staining. This is one of the most widely used staining methods in medical diagnosis and research. Hematoxylin stains cell nuclei a deep blue or purple color. It achieves this by binding to acidic components within the nucleus, such as nucleic acids and proteins, which have a negatively charged phosphate backbone. The precise mechanism of binding involves a mordant, usually a metallic salt like aluminum potassium sulfate (alum), which helps the dye adhere to cellular structures.

The preparation of the hematoxylin solution is critical for optimal staining results. It often involves a ripening or oxidation process to convert hematoxylin into its active staining form, hematein. Different formulations exist, including progressive and regressive staining methods, which dictate the duration of staining and subsequent rinsing steps.
