Halfsynthetic is a term sometimes used to describe substances produced from natural starting materials that are subsequently modified by chemical synthesis, placing them between fully natural products and fully synthetic compounds. It is closely related to the more common term semi-synthetic, and in many contexts the two words are used interchangeably. However, halfsynthetic is not standardized across disciplines and may be encountered inconsistently.

In pharmacology, many medicines are described as semi- or halfsynthetic; they begin with a natural scaffold such as a bacterial metabolite, plant alkaloid, or other natural product and are chemically altered to improve potency, spectrum, or pharmacokinetic properties. Classic examples include penicillin derivatives such as ampicillin and amoxicillin, which extend the penicillin family to broaden activity and stability. Opioids like diamorphine (heroin) are diacetylated derivatives of morphine and are often described as semi-synthetic. Artemisinin derivatives used in malaria treatment, such as artesunate, are also obtained by partial chemical modification of the natural product.

The halfsynthetic approach aims to preserve useful biological activity while enabling practical production and improved therapeutic characteristics. Benefits include leveraging nature’s diversity and reducing the need for entirely novel synthesis, but challenges include the complexity of modification, regulatory classification, variability of natural starting materials, and intellectual property considerations.

Because halfsynthetic is less standardized than semi-synthetic, it is prudent to verify usage within a given field and to prefer the latter term in formal scientific writing.
