Gproteiinikytköksellisiä, often abbreviated as GPCRs, represent a large and diverse family of cell surface receptors that play a crucial role in cellular signal transduction. These proteins are embedded within the cell membrane and are responsible for detecting a wide array of extracellular stimuli, including hormones, neurotransmitters, light, and odors. Upon binding of a specific ligand to the GPCR, the receptor undergoes a conformational change.

This conformational change activates an associated intracellular protein known as a G protein. G proteins are heterotrimeric proteins, consisting of alpha, beta, and gamma subunits. When a GPCR is activated, it facilitates the exchange of guanosine diphosphate (GDP) for guanosine triphosphate (GTP) on the alpha subunit of the G protein. This GTP binding event causes the dissociation of the alpha subunit from the beta-gamma dimer.

Both the activated alpha subunit (bound to GTP) and the beta-gamma dimer can then interact with and modulate the activity of various downstream effector proteins. These effectors include enzymes, such as adenylyl cyclase and phospholipase C, and ion channels. The activation or inhibition of these effectors leads to the generation of intracellular second messengers or changes in ion concentrations, ultimately triggering a specific cellular response. This cascade of events allows cells to respond to external signals in a precise and regulated manner. GPCRs are involved in a vast number of physiological processes and are thus targets for a significant proportion of modern pharmaceuticals.
