Glycanprotein, commonly referred to as a glycoprotein, is a protein that carries covalently attached carbohydrate chains, or glycans. Glycosylation, the attachment of these glycans, occurs predominantly in the secretory pathway and is essential for proper folding, stability, trafficking, and function of many proteins. The attached glycans can make up a substantial portion of the molecule’s mass and contribute to micro-heterogeneity among protein molecules produced by the same gene.

Two major classes are glycoproteins: N-linked and O-linked. In N-linked glycoproteins, glycans are attached to the amide nitrogen of asparagine within the consensus sequence Asn-X-Ser/Thr, where X is not proline. O-linked glycoproteins have glycans attached to the hydroxyl group of serine or threonine residues. There are additional, less common types of glycan linkages as well.

Biosynthesis and processing occur mainly in the endoplasmic reticulum and Golgi apparatus. Initial glycosylation in the ER assists protein folding and quality control, while further processing in the Golgi tailors glycan structures to influence localization, stability, and interactions with other molecules, including lectins.

Glycoproteins perform numerous roles, including mediating cell–cell and cell–matrix interactions, modulating immune responses, and protecting proteins from proteolysis. The glycans also influence solubility, half-life, and recognition by other biomolecules.

Medical relevance is significant. Defects in glycosylation cause congenital disorders of glycosylation. Many therapeutic proteins are glycoproteins, and their efficacy and pharmacokinetics depend on glycosylation patterns. Abnormal glycosylation is associated with diseases such as cancer and inflammation, and glycoengineering aims to optimize glycan structures for therapy. Examples include immunoglobulin G, transferrin, erythropoietin, and mucins.
