Phospho-ERK refers to the phosphorylated form of Extracellular signal-Regulated Kinase (ERK). ERK is a key protein in the mitogen-activated protein kinase (MAPK) signaling pathway, which plays a critical role in regulating cell proliferation, differentiation, survival, and migration. Phosphorylation is a crucial post-translational modification that activates or deactivates proteins. In the case of ERK, phosphorylation at specific threonine and tyrosine residues within its activation loop by upstream kinases, such as MEK (MAPK/ERK kinase), leads to its activation.

Activated phospho-ERK then translocates to the nucleus or remains in the cytoplasm, where it phosphorylates a wide range of downstream target proteins, including transcription factors, other kinases, and cytoskeletal proteins.
