Fosfatase is a broad term for enzymes that remove phosphate groups from molecules. This process is called dephosphorylation. These enzymes are crucial in many biological processes as they can activate or deactivate proteins, regulate metabolic pathways, and play a role in cell signaling.

There are many different types of phosphatases, often categorized by the type of bond they cleave within the phosphate ester. For example, alkaline phosphatase can remove phosphate groups from a wide range of substrates at alkaline pH levels, while acid phosphatase functions optimally at acidic pH. The activity of phosphatases is tightly regulated. Dysregulation of phosphatase activity has been implicated in various diseases, including cancer, neurological disorders, and metabolic diseases. For instance, certain tyrosine phosphatases are considered tumor suppressors as they can counteract the effects of oncogenic tyrosine kinases. Conversely, the overexpression or hyperactivity of some phosphatases can also drive disease progression. Therefore, phosphatases are significant targets for drug development in a variety of therapeutic areas.
