Enzymecoupled refers to systems in which one enzymatic reaction is linked to another reaction, process, or detection step so that the product of the first enzyme is consumed or transformed by a second enzyme. The coupling creates an enzymatic cascade in which an initial substrate event is translated into a measurable signal or into a final product that can be readily monitored.

In laboratory practice, enzyme-coupled assays use a primary enzyme reaction whose product is processed by a secondary enzyme. This arrangement enables readouts such as colorimetric, fluorometric, luminescent, or electrochemical signals. Classic examples include glucose assays that couple glucose oxidase to horseradish peroxidase, where hydrogen peroxide produced by the first enzyme drives a color change; and lactate or pyruvate assays that rely on cofactors like NADH/NADPH in a coupled dehydrogenase reaction. ATP-based assays often use luciferase to generate light from ATP produced in the primary reaction.

Applications of enzymecoupled strategies span clinical diagnostics, food and environmental testing, and biosensor design. They enable increased sensitivity, broader dynamic range, or compatibility with standard detectors. In biochemical research, enzyme coupling helps quantify low-abundance substrates or monitor metabolic flux through a pathway.

Key considerations include ensuring efficient coupling and minimal side reactions, maintaining cofactor recycling, and addressing stability and diffusion limitations.
