Emäsparisääntöjen, often translated as base pairing rules, are fundamental principles in molecular biology that describe the specific way nitrogenous bases in nucleic acids (DNA and RNA) associate with each other. These rules are crucial for the structure and function of these genetic molecules.

In DNA, the base pairing is always between adenine (A) and thymine (T), forming a hydrogen bond between them, and between guanine (G) and cytosine (C), also forming hydrogen bonds. This A-T and G-C pairing is known as complementary base pairing. Adenine and guanine are purines, which have a double-ring structure, while thymine and cytosine are pyrimidines, with a single-ring structure. The pairing always involves one purine with one pyrimidine, maintaining a consistent width of the DNA double helix.

In RNA, the base pairing is similar, but with one key difference. While guanine still pairs with cytosine (G-C), adenine (A) pairs with uracil (U) instead of thymine (T). This A-U pairing is also mediated by hydrogen bonds. The presence of uracil in RNA and thymine in DNA is one of the distinguishing features between the two nucleic acids.
