Cell-cell adhesion structures are specialized protein complexes that mediate the attachment between adjacent cells or between cells and the extracellular matrix. These structures are crucial for maintaining tissue integrity, enabling cell migration, facilitating communication, and organizing cellular architecture. They play vital roles in embryonic development, wound healing, and immune responses.

There are several major types of cell-cell adhesion structures. Adherens junctions, desmosomes, and adherens junctions connect cells to each other by linking their cytoskeletons, typically actin filaments or intermediate filaments. These junctions involve transmembrane proteins called cadherins or desmosomal cadherins, which bind to similar proteins on neighboring cells. Intracellularly, these cadherins are anchored to the cytoskeleton via adapter proteins. Tight junctions, also known as occluding junctions, form seals between epithelial cells, preventing the passage of molecules through the paracellular space and maintaining cell polarity. They are composed of claudins and occludins, transmembrane proteins that interact with each other to create a continuous belt around the cell. Gap junctions, on the other hand, facilitate direct communication between cells by forming channels that allow small molecules and ions to pass from one cytoplasm to another. These channels are formed by connexins or innexins.

Cell-cell adhesion structures are dynamic and their formation and disassembly are tightly regulated by intracellular signaling pathways. Dysregulation of these structures can lead to various diseases, including cancer, autoimmune disorders, and developmental abnormalities. Understanding the molecular mechanisms underlying cell-cell adhesion is fundamental to comprehending tissue organization and developing therapeutic strategies for related pathologies.
