Calcineurmediated refers to processes or effects that are controlled or influenced by calcineurin. Calcineurin is a calcium- and calmodulin-dependent protein phosphatase that plays a crucial role in various cellular signaling pathways. Its primary function involves dephosphorylating target proteins, thereby altering their activity and downstream effects.

One of the most well-studied calcineurmediated pathways involves the activation of T lymphocytes. Following T cell receptor stimulation, an increase in intracellular calcium triggers calcineurin activity. Activated calcineurin then dephosphorylates the transcription factor NFAT (nuclear factor of activated T-cells), allowing it to translocate to the nucleus and promote the expression of genes essential for T cell activation, such as interleukin-2. This calcineurmediated pathway is a key target for immunosuppressive drugs like cyclosporine and tacrolimus, which inhibit calcineurin activity to prevent transplant rejection and treat autoimmune diseases.

Beyond immunology, calcineurmediated signaling is implicated in other cellular processes. This includes neuronal function, where calcineurin influences synaptic plasticity, learning, and memory. It is also involved in cardiac hypertrophy and skeletal muscle development.
