Antibody-independent is an adjective used to describe processes, responses, or techniques that do not require antibodies to function or be measured. The term is common in immunology, diagnostics, and biotechnology to distinguish from antibody-dependent approaches that use antibodies as reagents or effectors.

In immunology, antibody-independent immunity encompasses innate and cell-mediated responses that do not rely on antibodies. Examples include phagocytosis by macrophages driven by pattern-recognition receptors, natural killer cell cytotoxicity not mediated by antibodies, and T-cell–mediated responses recognizing antigens via MHC molecules. The complement system can also be activated through antibody-independent pathways, such as the lectin and alternative pathways, while antibodies can modulate some downstream effects.

In diagnostics and research, antibody-independent methods detect pathogens, biomarkers, or molecules without using antibodies. Common approaches include nucleic acid amplification tests (PCR, RT-PCR), sequencing-based detection, culture methods, and mass spectrometry-based proteomics. Aptamer-based sensors and other ligand-based assays provide antibody-free alternatives to immunoassays.

In therapeutics, antibody-independent strategies rely on small molecules, nucleic acids, or cell-based therapies rather than monoclonal antibodies. Examples include antiviral small-molecule drugs, kinase inhibitors, CAR-T therapies, and innate immune modulators. These approaches can be advantageous when antigen variability or tissue penetration limits antibody-based options.
