Amidbindning is the Danish term for the process of forming an amide bond, also known as amidation. The reaction joins a carboxylic acid with an amine (primary or secondary) to produce an amide and a molecule of water or another small leaving group such as a carbamate. In industrial chemistry and pharmaceutical synthesis, amidbinding is fundamental for constructing peptide chains, drugs containing amide linkages, and various polymer backbones.

The reaction typically proceeds through activation of the carboxylic acid. Common activation methods include the use of carbodiimides (e.g., DCC or EDC), acid chlorides, or mixed anhydrides. When the activated acid encounters the amine nucleophile, the nitrogen attacks the carbonyl carbon, leading to an intermediate tetrahedral adduct that collapses to release the amide. In aqueous media, carbodiimide activation is often combined with a coupling additive such as N-hydroxysuccinimide to suppress side reactions and improve yields.

Amidbinding plays a key role in peptide synthesis. In solid‑phase peptide synthesis (SPPS) the amide bond is formed repeatedly between a resin‑bound amino acid and the next residue, enabling rapid assembly of long chains. The reaction is also employed in the synthesis of small‑molecule pharmaceuticals where an amide linkage confers metabolic stability and favorable pharmacokinetics.

Environmental and safety considerations arise from the generation of urea byproducts in carbodiimide reactions and from the use of volatile organophosphorus chlorides. Accordingly, green chemistry approaches explore enzymatic amidation, flow‑chemistry setups, and recyclable activation systems.
