An aldol reaction is a fundamental carbon-carbon bond-forming reaction in organic chemistry. It involves the addition of an enol or enolate ion to a carbonyl compound, such as an aldehyde or ketone. This reaction is named after the fact that the product is a beta-hydroxy aldehyde or ketone, which contains both an aldehyde/ketone functional group and an alcohol functional group. The aldol reaction can occur under either acidic or basic conditions.

Under basic conditions, a base abstracts an alpha-hydrogen from a carbonyl compound, forming a nucleophilic enolate ion. This enolate then attacks the electrophilic carbonyl carbon of another molecule of the same or a different aldehyde or ketone. The resulting alkoxide is then protonated to yield the beta-hydroxy carbonyl compound. If the starting carbonyl compound has no alpha-hydrogens, it cannot form an enolate and thus cannot act as the nucleophile.

Under acidic conditions, an acid catalyst protonates the carbonyl oxygen of one molecule, making its alpha-carbon more acidic. This allows for enolization, where a proton is transferred to the alpha-carbon, forming an enol. The enol, acting as a nucleophile, then attacks the protonated carbonyl carbon of another molecule. Subsequent deprotonation yields the beta-hydroxy carbonyl product.

The aldol reaction can be followed by a dehydration step, known as the aldol condensation, which occurs readily when the beta-hydroxy carbonyl compound is heated or treated with acid or base. This dehydration removes a molecule of water, forming an alpha,beta-unsaturated aldehyde or ketone. This condensation step is often favored due to the increased stability of the conjugated product. The aldol reaction and condensation are versatile tools for building more complex organic molecules.
