YACvektoreita

YACvektoreita is a Finnish term that translates to "YAC vectors" or "YAC cloning vectors" in English. These are specialized DNA molecules used in molecular biology, particularly in the field of genetic engineering and cloning. YAC stands for Yeast Artificial Chromosome. Yeast artificial chromosomes are designed to replicate and be maintained within yeast cells, similar to how natural chromosomes are.

The primary function of YAC vectors is to clone very large fragments of DNA. Unlike traditional cloning vectors such as plasmids or bacteriophages, which have limitations on the size of DNA they can accommodate, YAC vectors can carry inserts of hundreds of thousands to even millions of base pairs. This makes them invaluable for cloning large genomic regions, such as entire genes, gene clusters, or even chromosomal arms.

The construction of a YAC vector involves several key components: an origin of replication, centromere sequences, and telomere sequences, all derived from yeast chromosomes. These elements allow the vector to behave like a miniature chromosome within the yeast host. Foreign DNA of interest is ligated between the arms of the YAC vector, and this recombinant molecule is then introduced into yeast cells through a process called transformation.

Once inside the yeast, the YAC vector, along with its inserted DNA fragment, replicates and segregates during cell division, ensuring that the cloned DNA is stably maintained and passed on to daughter cells. This makes YACs useful for constructing genomic libraries, studying gene regulation over large distances, and for applications in genomics and gene therapy where large DNA segments are required. Despite the development of newer technologies, YACs remain a significant tool for cloning and manipulating large DNA sequences.
