Vävnadsfaktorerna, also known as tissue factors, are a group of proteins that play a crucial role in the blood coagulation cascade, specifically in initiating the extrinsic pathway. The most prominent and well-studied of these is Tissue Factor Pathway Inhibitor (TFPI), although the term can sometimes encompass other related proteins involved in coagulation regulation. TFPI is a potent inhibitor of the tissue factor (TF)-factor VIIa complex, which is the primary initiator of the extrinsic pathway.

The primary function of TFPI is to prevent the excessive or inappropriate formation of blood clots. When tissue damage occurs, tissue factor is exposed to the bloodstream, leading to the activation of factor VII. This TF-factor VIIa complex then activates factor X, leading to the generation of thrombin and subsequent clot formation. TFPI circulates in the blood and binds to the TF-factor VIIa complex, thereby inhibiting its activity and limiting the extent of coagulation.

There are different isoforms of TFPI, each with slightly varying structures and mechanisms of action, but all contribute to the delicate balance of hemostasis. Dysfunction or deficiency in TFPI can lead to an increased risk of bleeding disorders, while excessive TFPI activity might predispose individuals to thrombotic events.
