Twoenzyme

Twoenzyme is a term used in biocatalysis and synthetic biology to describe systems that harness two enzymatic activities for a coordinated transformation. The concept encompasses both bifunctional enzymes in which two catalytic domains reside within a single polypeptide chain and cascaded sets of two enzymes that operate in sequence, either in vitro or inside living cells.

In bifunctional twoenzyme constructs, domain architecture is arranged to support substrate channeling and to reduce diffusion losses. The linker length and flexibility between domains influence activity, temperature stability, and potential regulation. In cascaded twoenzyme systems, the product of the first enzyme serves as the substrate for the second, enabling multi-step transformations with improved overall throughput compared with single-step reactions. Spatial organization, cofactor reuse, and microenvironment control can further enhance efficiency.

Twoenzyme designs are used to enable rapid synthesis of complex molecules, renewable cofactor regeneration, and compact biosensors. They are common in metabolic engineering, where pathway bottlenecks can be alleviated by integrating two enzymatic steps, and in protein engineering, where fusion strategies aim to create self-contained catalytic modules.

Challenges include maintaining balanced expression levels, avoiding steric clashes between domains, and ensuring proper folding. Natural two-enzyme systems, such as certain multi-domain synthases, demonstrate the potential of intrinsic substrate channeling, while engineered twoenzyme constructs illustrate the feasibility of modular biocatalysis.
