TurnoverFrequency

TurnoverFrequency is a measure of catalytic activity, typically defined as the number of substrate molecules converted to product per catalytic site per unit time. It is commonly abbreviated as TOF and is most often expressed in reciprocal seconds (s^-1). In enzymology and catalysis, TOF describes how rapidly a single active site cycles through catalytic turnovers under specified conditions.

TOF is related to but distinct from turnover number (TON). TON denotes the total number of substrate molecules converted per active site before the catalyst becomes inactive, whereas TOF refers to the rate of turnover, either at a given moment or averaged over a period. In enzymes, TOF at saturating substrate concentration is often denoted kcat, the maximum catalytic rate per active site.

Calculation of TOF requires the rate of product formation and the number of active sites. A common expression is TOF = rate / [E], where rate is the amount of product formed per unit time and [E] is the concentration of active catalytic sites. For heterogeneous catalysts, TOF is typically calculated per active site on the surface or per defined catalytic center.

Factors that influence TOF include substrate concentration, temperature, pH (particularly for enzymes), presence of inhibitors or activators, co-factors, and the physical state of the catalyst. TOF can vary with substrate concentration, and in enzymes, kcat represents the intrinsic maximum TOF under saturating conditions, independent of binding affinity.
