Triphosphatedependent refers to a biological process or enzyme activity that requires the presence of nucleoside triphosphates, most commonly adenosine triphosphate (ATP) or guanosine triphosphate (GTP), to function. These high-energy molecules serve as an energy source or a substrate for the reaction. Many cellular processes rely on the hydrolysis of the phosphate bonds in triphosphates to drive them forward.

Enzymes that are triphosphatedependent are often involved in fundamental biological functions. For instance, DNA and RNA polymerases, which synthesize nucleic acids, are triphosphatedependent as they utilize nucleoside triphosphates as building blocks and the energy released from pyrophosphate cleavage fuels the polymerization process. Motor proteins, such as myosin and kinesin, also exhibit triphosphatedependent activity, using ATP hydrolysis to generate the mechanical force needed for movement along cellular tracks.

Other examples include various signaling pathways where GTP-binding proteins (G proteins) are activated by GTP binding, regulating downstream effectors. Kinases, a large class of enzymes, catalyze the transfer of a phosphate group from ATP to a substrate, and are thus inherently triphosphatedependent. The study of triphosphatedependent mechanisms is crucial for understanding cellular energy metabolism, signal transduction, and molecular machinery.
