TLRIL1signaalin refers to the signaling pathway initiated by Toll-like receptor 1 (TLR1). Toll-like receptors are a class of proteins that play a key role in the innate immune system. They recognize conserved molecular patterns associated with pathogens, known as pathogen-associated molecular patterns (PAMPs), and also damage-associated molecular patterns (DAMPs) released by stressed or dying host cells.

When TLR1 binds to its specific ligand, typically a triacylated lipopeptide found on the surface of certain bacteria, it forms a heterodimer with either TLR2 or TLR6. This complex then recruits an adapter protein called MyD88 (myeloid differentiation primary response 88). MyD88 acts as a scaffold, bringing together downstream signaling molecules.

The activation of MyD88 leads to the activation of a kinase cascade involving IRAKs (interleukin-1 receptor-associated kinases) and TRAF6 (TNF receptor-associated factor 6). This cascade ultimately results in the activation of transcription factors, primarily NF-κB (nuclear factor kappa-light-chain-enhancer of activated B cells) and AP-1 (activator protein 1). These transcription factors then translocate to the nucleus and induce the expression of pro-inflammatory cytokines, chemokines, and other immune mediators. These molecules help to recruit immune cells to the site of infection, initiate an inflammatory response, and ultimately combat the pathogen. TLR1 signaling is therefore crucial for mounting an effective innate immune defense against certain bacterial infections.
