The well was discovered on January 10, 1901, by the Lewis Ranch, a partnership between George Mitchell and his brother-in-law, Leon "Pappy" Gladin. The discovery marked the beginning of the modern oil industry in the United States. The well was named "Spindletop" due to the distinctive shape of the land where it was found, and "Gladys" was named after the daughter of George Mitchell.

The Spindletop Gladys well was the first in the world to produce oil at a rate of over 100 barrels per day. This discovery led to a significant increase in oil production and the establishment of the oil industry in the United States. The well was operated by the Texas Company, which was later acquired by Standard Oil. The well was eventually plugged and abandoned in 1915.

The Spindletop Oil Field is now a historic site and is listed on the National Register of Historic Places.
