Sfingolipidoses
Sfingolipidoses are a group of rare genetic metabolic disorders that affect sphingolipid metabolism. Sphingolipids are a class of lipids that are important components of cell membranes, particularly in the nervous system. These disorders arise from inherited deficiencies in specific enzymes that are responsible for breaking down or recycling sphingolipids. When these enzymes are not functioning correctly, sphingolipids accumulate to toxic levels within cells, primarily in the lysosomes. This accumulation leads to progressive cellular damage and dysfunction, particularly in nerve cells, but also affecting other organs like the liver, spleen, and bone marrow.
The clinical presentation of sfingolipidoses varies widely depending on the specific enzyme deficiency and the type
