SYCP3

SYCP3, or synaptonemal complex protein 3, is a protein encoded by the SYCP3 gene in humans. It is a structural component of the synaptonemal complex, a proteinaceous scaffold formed between homologous chromosomes during meiotic prophase I.

During meiosis, SYCP3 localizes to the lateral elements of the synaptonemal complex and contributes to chromosome pairing, synapsis, and recombination. It is typically expressed in germ cells undergoing meiosis, with highest levels in spermatocytes; expression in most somatic tissues is low or absent.

Genetic and cellular studies in model organisms indicate that SYCP3 plays an essential role in meiotic chromosome synapsis. Loss of SYCP3 disrupts synapsis and can lead to meiotic arrest and infertility, underscoring its importance for proper chromosome segregation during gametogenesis.

Clinical relevance: In humans, SYCP3 expression is normally restricted to testis, but aberrant expression has been observed in various cancers, where it is classified as a cancer/testis antigen and explored as a target for immunotherapy. The meiotic origin of the protein also makes it a subject of research into mechanisms of chromosomal instability when misexpressed in somatic cells.

Structure and interactions: SYCP3 is predicted to be a coiled-coil protein and can form higher-order assemblies. It interacts with other components of the synaptonemal complex, including SYCP1 and SYCP2, contributing to the assembly and stability of the complex during meiosis.
