Membership in a receptor family is typically determined by gene homology, meaning the genes encoding these receptors have a shared evolutionary origin. This shared ancestry often translates to conserved structural motifs, particularly within the ligand-binding domain and the transmembrane regions, as well as similar signal transduction mechanisms.

The classification of receptors into families helps researchers predict the function of newly discovered receptors based on the known properties of their relatives. For example, the G protein-coupled receptor (GPCR) superfamily is one of the largest and most diverse families, characterized by a seven-transmembrane helix structure. They are involved in a vast array of physiological processes, from neurotransmission to hormone signaling.
