Metalloproteinases1

Metalloproteinases1, also referred to in the literature as MMP‑1 or collagenase‑1, belongs to the metalloproteinase family of zinc‑dependent endopeptidases. These enzymes hydrolyse peptide bonds within the extracellular matrix (ECM) and are crucial for normal tissue remodeling, wound healing, and embryonic development. The protein is synthesised as a preproenzyme and released into the extracellular space where it undergoes proteolytic activation.

In human cells, the MMP1 gene is located on chromosome 11q22.2 and encodes a secreted protein of about 57 kDa. The enzyme preferentially cleaves native fibrillar collagens type I, II and III, a capability that facilitates cell migration through collagenous tissues. Metalloproteinases1 is regulated at multiple levels: transcriptional control by cytokines such as interleukin‑1β and tumor necrosis factor‑α, post‑translational activation by furin‑like convertases, and inhibition by tissue inhibitors of metalloproteinases (TIMPs), primarily TIMP‑1 and TIMP‑2.

The enzymatic activity of METALLOPROTEINASES1 is implicated in several pathological processes. Elevated expression has been observed in arthritis, where it contributes to cartilage degradation; in cardiovascular disease, through remodeling of the vascular wall; and in various cancers, where it facilitates tumour invasion and metastasis by breaking down basement membranes. Because of its central role in ECM turnover, MMP‑1 is a target for therapeutic agents such as selective inhibitors, although clinical success has been limited by issues of specificity and side effects.

Research continues to explore the structural basis of substrate recognition and the development of more selective inhibitors. The study of metalloproteinases1 thus remains a vital area for understanding both normal physiology and the molecular mechanisms underlying numerous human diseases.
