Megakarüotsütopoiesis

Megakarüotsütopoiesis is the biological process by which megakaryocytes, large multinucleated cells found in the bone marrow, are formed. This complex developmental pathway originates from multipotent hematopoietic stem cells, which differentiate through a series of progenitor stages. Key intermediate cell types include megakaryocyte progenitor cells (Mk-Ps) and colony-forming unit-megakaryocytes (CFU-Meg). Throughout this process, cells undergo significant changes in size, nuclear content, and cytoplasmic characteristics.

The development of megakaryocytes is driven by a network of growth factors and cytokines, with thrombopoietin (TPO) being the primary regulator. TPO binds to its receptor, c-Mpl, on hematopoietic stem cells and progenitor cells, initiating signaling cascades that promote their proliferation, differentiation, and survival. Other factors such as interleukin-3 (IL-3) and stem cell factor (SCF) can also play supporting roles.

As megakaryocytes mature, they exhibit endomitosis, a unique form of cell division where DNA replicates but the cell does not divide. This results in the formation of polyploid nuclei, with some megakaryocytes reaching ploidy levels of 64N or higher. The cytoplasm of these cells becomes increasingly granular and basophilic, storing essential proteins and molecules required for platelet formation. The final stage of megakarüotsütopoiesis involves the physical fragmentation of the megakaryocyte's long cytoplasmic extensions, called proplatelets, into numerous anucleated platelets, which are then released into the bloodstream. This intricate process is crucial for maintaining adequate platelet counts, essential for hemostasis and blood clotting.
