MYOM1

MYOM1 is a human gene that encodes myomesin-1, a structural protein of the sarcomere’s M-band in striated muscle. Myomesin-1 is part of the myomesin family and localizes to the central region of the sarcomere, where it contributes to the architecture and mechanical stability of the M-band.

Functionally, myomesin-1 helps organize and anchor the thick filaments by crosslinking myosin to the titin filament network at the M-band. This crosslinking supports the alignment and integrity of the sarcomere during contraction and helps maintain proper spacing between thick filaments. Myomesin-1 participates in sarcomere assembly during development and is implicated in the structural response to mechanical stress. The protein interacts with titin and myosin, and it associates with other M-band components to sustain the cooperative function of the sarcomere.

Structural features of myomesin-1 include immunoglobulin-like and fibronectin type III domains, and the protein can form dimers that contribute to the organization of the M-band. The MYOM1 gene shows expression in cardiac muscle and skeletal muscle, with multiple isoforms generated by alternative splicing, reflecting tissue-specific and developmental regulation.

Clinical significance regarding MYOM1 is an area of ongoing research. Variants in MYOM1 have been reported in association with cardiomyopathies in some studies, including dilated cardiomyopathy, but the pathogenic role of specific variants remains to be fully established. Additional research is needed to clarify MYOM1’s contribution to muscle disease and sarcomere function.
