MScleavable

MScleavable is a term used in mass spectrometry to describe molecules or components within a larger structure that are designed to break apart predictably under specific mass spectrometry conditions, typically electrospray ionization (ESI) or matrix-assisted laser desorption/ionization (MALDI). This controlled fragmentation allows for the analysis of complex mixtures or the confirmation of specific molecular architectures.

The concept of MScleavable is often employed in the design of linker molecules used in chemical biology, proteomics, and drug delivery systems. These linkers can attach two or more entities, such as a protein to a small molecule, or two peptides. The MScleavable linker is engineered so that upon ionization and subsequent fragmentation in the mass spectrometer, the linker breaks at a predetermined bond, releasing the attached components. This release can be detected and quantified, providing valuable information about the interaction or proximity of the original entities.

The design of MScleavable linkages relies on understanding the fragmentation patterns of specific chemical bonds within the mass spectrometer. Common motifs include disulfide bonds, ester bonds, or specific peptide sequences that are susceptible to cleavage by collision-induced dissociation (CID) or other fragmentation techniques. The advantage of using MScleavable elements is that they enable the unambiguous identification and characterization of analytes that might otherwise be difficult to analyze due to their size or complexity.
