Kryoprotektantteina

Kryoprotektantteina, or cryoprotectant agents, are substances used to protect biological material from damage caused by freezing and ice formation. They help preserve the structure and function of cells, tissues, and other biological samples during cryopreservation by reducing ice crystal formation, lowering freezing points, and stabilizing membranes and proteins. Cryoprotectants can act inside cells (permeating) or outside cells (non-permeating), and are often used in combinations to balance efficacy and toxicity.

The protective mechanisms of cryoprotectants include reducing intracellular ice formation through colligative effects, promoting vitrification (glass-like solidification) instead of crystal growth, and stabilizing cellular membranes, enzymes, and macromolecules by replacing water in hydration shells. Permeating cryoprotectants enter cells and help minimize intracellular ice, while non-permeating agents protect the extracellular environment and control osmotic shifts during cooling and warming.

Common examples include permeating agents such as glycerol, dimethyl sulfoxide (DMSO), ethylene glycol, and propylene glycol. Non-permeating agents often include sugars like sucrose and trehalose, polyols, and certain polymers. The choice of cryoprotectant depends on the type of sample, the cooling and warming rates, and the acceptable level of toxicity.

Applications are broad and include cryopreservation of cells, embryos, oocytes, sperm, and tissues for medical research and assisted reproduction, as well as various industrial uses in the food and biotechnology sectors. Safety and handling are important, as many cryoprotectants can be toxic at high concentrations and require careful optimization and post-thaw removal or dilution.
