IL1IL18signaation refers to the complex signaling pathways involving interleukin-1 (IL-1) and interleukin-18 (IL-18). These are pleiotropic cytokines, meaning they have diverse effects on various cell types and play crucial roles in both innate and adaptive immunity. IL-1 and IL-18 are primarily produced by immune cells, such as macrophages and dendritic cells, in response to pathogens or tissue damage. Their signaling often occurs through distinct but sometimes overlapping receptor complexes on target cells.

The IL-1 signaling pathway is initiated when IL-1 binds to its receptor IL-1R1, leading to the recruitment of adaptor proteins and activation of downstream kinases, most notably the NF-κB pathway. This activation results in the transcription of genes involved in inflammation, cell survival, and proliferation. IL-18 also signals through its receptor IL-18R1, often in conjunction with a co-receptor, IL-18RAP. This binding activates intracellular signaling cascades, including MyD88-dependent pathways and the induction of NF-κB and MAPK signaling.

Interestingly, IL-1 and IL-18 signaling can be synergistic. For instance, IL-1 can prime cells to respond more robustly to IL-18. This interplay is particularly important in the activation of T helper 1 (Th1) cells and the production of interferon-gamma (IFN-γ). Both cytokines are implicated in a wide range of physiological and pathological processes, including host defense against infections, inflammatory diseases, autoimmune disorders, and cancer. Dysregulation of IL-1 and IL-18 signaling contributes to the pathogenesis of numerous diseases.
