GlcN6phosphate

GlcN6phosphate, or glucosamine‑6‑phosphate, is a monosaccharide derivative that contains an amino group at the second carbon and a phosphate group at the sixth carbon. Its chemical formula is C6H13NO9P and it exists as a zwitterion at physiological pH. The compound is a pivotal intermediate in the hexosamine biosynthetic pathway, which supplies the core monosaccharides for glycoprotein and glycolipid synthesis.

During the pathway, fructose‑6‑phosphate is conjugated with glutamine by glutamine:fructose‑6‑phosphate amidotransferase, forming glucosamine‑6‑phosphate and glutamate. GlcN6phosphate is then epimerised and subsequently deaminated to yield fructose‑6‑phosphate, thereby linking amino‑sugar metabolism with glycolysis. Glycosylation of proteins and lipids requires UDP‑GlcNAc, which is produced from GlcN6phosphate via a series of enzymatic steps. In plants, GlcN6phosphate can act as a signaling molecule that regulates transcription of stress‑response genes.

Because of its centrality in nitrogen flux and carbohydrate metabolism, deregulation of GlcN6phosphate levels can affect cell growth, stress responses and disease states such as cancer and diabetes. The compound is therefore a target of interest in metabolic research and drug development.
