GalNActerminated

GalNActerminated refers to glycans or glycoproteins in which the terminal sugar residue is N-acetylgalactosamine (GalNAc). In this context, the non-reducing end of the oligosaccharide presents GalNAc, and the structure may be referred to as GalNAc-terminated or GalNAc-terminated O-glycans. A canonical example is the Tn antigen, GalNAcα-O-Ser/Thr, which represents the initial product of mucin-type O-glycosylation before further extension or elongation.

Biosynthesis and contexts: GalNAc is typically attached to serine or threonine residues by polypeptide N-acetylgalactosaminyltransferases (ppGalNAc-Ts) during O-glycosylation. If subsequent glycan elaboration is limited or altered, the resulting product can remain GalNAc-terminated. GalNAc-terminated glycans are common on mucins and certain glycoproteins, and they are also a component of blood group A antigens in the appropriate disaccharide context (GalNAcα1-3Gal).

Biological and clinical relevance: GalNAc-terminated structures can serve as epitopes for specific lectins and antibodies. The exposure of GalNAc-terminated glycans, such as the Tn antigen, is associated with various pathological states, including cancer, where truncated O-glycosylation patterns occur. Because of their distinct surface appearance, GalNAc-terminated glycans are studied as biomarkers and potential targets for diagnostics or vaccines.

Detection and analysis: Detection commonly employs lectins that specifically bind GalNAc, such as Helix pomatia agglutinin (HPA) or Vicia villosa agglutinin (VVA), as well as mass spectrometry and glycan microarrays to characterize terminal GalNAc-containing structures.
