This enzyme is primarily found in lysosomes, which are compartments within cells that break down waste materials. Glucocerebrosidase plays a crucial role in metabolizing certain fats and lipids.

Mutations in the GBA gene can lead to a deficiency or malfunction of the glucocerebrosidase enzyme. When the enzyme is not functioning correctly, these specific lipids can accumulate to toxic levels within cells, particularly in macrophages. This accumulation can cause a range of health problems.

The most well-known condition associated with GBA mutations is Gaucher disease. Gaucher disease is a rare genetic disorder characterized by the buildup of glucocerebroside in various organs, including the spleen, liver, bone marrow, and lungs. The severity and specific symptoms of Gaucher disease can vary widely depending on the type of GBA mutation and the residual enzyme activity. Symptoms can include enlarged spleen and liver, anemia, bone problems, and neurological issues.

Other conditions, such as Parkinson's disease, have also been linked to GBA mutations. While not everyone with a GBA mutation will develop Parkinson's disease, studies suggest that certain GBA variants may increase an individual's risk of developing this neurodegenerative disorder. Research is ongoing to fully understand the complex relationship between GBA gene mutations and neurological conditions. Genetic testing can identify specific GBA mutations, which can be important for diagnosis, understanding disease risk, and in some cases, for guiding treatment options.
