Cysteineetikettering, often referred to as cysteine labeling, is a technique used in molecular biology and biochemistry to selectively attach a label or tag to cysteine residues within a protein or peptide. Cysteine is an amino acid characterized by its thiol side chain (-SH), which is highly reactive and can participate in disulfide bond formation or undergo various chemical modifications. This reactivity makes cysteine a prime target for selective labeling strategies.

The primary goal of cysteine labeling is to introduce a functional moiety, such as a fluorescent dye, a biotin tag, a radioactive isotope, or a cross-linking agent, onto a specific cysteine residue. This introduced label can then be utilized for a variety of downstream applications. For instance, fluorescent labels allow for the visualization and tracking of proteins in cellular environments using microscopy or flow cytometry. Biotin tags enable affinity purification of the labeled protein using streptavidin-coated beads. Radioactive labels are useful for quantifying protein levels or studying metabolic pathways. Cross-linking agents can be used to study protein-protein interactions.

Several methods exist for cysteine labeling. One common approach involves maleimide chemistry, where the thiol group of cysteine reacts with the electrophilic double bond of maleimide-derivatized labels. Another method utilizes iodoacetamide, which alkylates the thiol group. Thiol-disulfide exchange reactions can also be employed, where a labeled molecule with a free thiol displaces the hydrogen atom of the cysteine thiol. The choice of labeling method often depends on the specific label desired, the protein of interest, and the experimental conditions. This technique is crucial for protein characterization, drug discovery, and understanding biological processes.
