Cdh1mediated

Cdh1mediated is often encountered as shorthand for processes driven by Cdh1, a name that appears in two distinct protein contexts in animals. The two proteins are unrelated despite the similar name: Cdh1, an activator of the anaphase-promoting complex/cyclosome (APC/C) in cell-cycle control, and CDH1, the gene encoding E-cadherin, a key component of epithelial adherens junctions.

Cdh1 in APC/C: Cdh1 (APC/Cdh1) is a regulatory co-activator of the APC/C E3 ubiquitin ligase. It activates APC/C from late mitosis through G1, promoting ubiquitination and proteasomal degradation of substrates such as cyclin B, cyclin A, securin, and geminin. This degradation enables mitotic exit and prevents premature entry into S phase. Cdh1 activity is controlled by phosphorylation by Cdks, nuclear localization, and inhibitors; its inactivation at the G1/S transition allows accumulation of cyclins necessary for S-phase entry.

CDH1/E-cadherin and Cdh1-mediated adhesion: CDH1 encodes E-cadherin, a calcium-dependent transmembrane glycoprotein that mediates homophilic cell–cell adhesion in epithelial tissues. E-cadherin forms adherens junctions by linking to the actin cytoskeleton via catenins. Loss or dysfunction of E-cadherin reduces cell adhesion and can promote epithelial-to-mesenchymal transition and cancer metastasis. Regulation occurs at transcriptional level by factors such as Snail and ZEB, and by promoter methylation and internalization. E-cadherin also participates in signaling through β-catenin dynamics.
