Calcium-binding proteins are a diverse group of proteins that have the ability to bind calcium ions (Ca2+). This interaction is crucial for numerous biological processes. These proteins are found throughout the body, with particularly important roles in cells.

The binding of calcium often induces a conformational change in the protein's structure. This alteration acts like a molecular switch, allowing the protein to activate or deactivate specific cellular functions. A classic example is calmodulin, which, upon binding calcium, can regulate the activity of various enzymes and ion channels.

Different calcium-binding proteins possess specialized domains that facilitate calcium binding. The EF-hand is one of the most common structural motifs, consisting of a helix-loop-helix structure that forms a pocket ideal for a single calcium ion. Proteins like calmodulin and troponin C contain multiple EF-hand domains.

The functions of these proteins are extensive. They are essential for intracellular calcium signaling, where they help translate changes in calcium concentration into cellular responses. Other key roles include buffering calcium levels to maintain homeostasis, as seen in calbindin, and facilitating muscle contraction, a function performed by troponin C in skeletal and cardiac muscle. They are also involved in processes like blood clotting, bone formation, and neurotransmitter release.
