BMPsignalering refers to the process by which Bone Morphogenetic Proteins (BMPs) communicate signals within and between cells. BMPs are a group of secreted proteins that play crucial roles in embryonic development, tissue repair, and differentiation of various cell types. The signaling pathway initiated by BMPs is a fundamental mechanism for controlling cell fate and patterning.

The BMP signaling pathway typically involves a BMP ligand binding to a heteromeric complex of type I and type II serine/threonine kinase receptors on the cell surface. This binding event activates the receptor complex, specifically the type I receptor, through phosphorylation by the type II receptor. The activated type I receptor then phosphorylates downstream intracellular signaling molecules, most notably the Smad proteins.

Activated Smads, particularly the receptor-regulated Smads (R-Smads) like Smad1, Smad5, and Smad8, translocate to the nucleus. There, they can interact with other transcription factors, co-activators, and co-repressors to regulate the expression of target genes. These target genes are involved in a wide range of cellular processes, including bone formation, cartilage development, cell proliferation, apoptosis, and epithelial-mesenchymal transitions.

Dysregulation of BMP signaling has been implicated in various diseases, including cancer, fibrosis, and developmental disorders.
