ARFmediated

ARFmediated refers to processes or events that are regulated or influenced by ADP-ribosylation factors (ARFs). ARFs are a family of small GTP-binding proteins that act as molecular switches in cellular signaling pathways. They are crucial for a variety of cellular functions, including vesicular transport, actin cytoskeleton organization, and lipid metabolism.

The activity of ARFs is tightly regulated by their binding to GTP or GDP. When bound to GTP, ARFs are in their active state and can recruit effector proteins to specific cellular locations, initiating downstream signaling cascades. Conversely, when bound to GDP, ARFs are inactive. This GTP/GDP cycling is controlled by guanine nucleotide exchange factors (GEFs), which promote GTP binding and activation, and GTPase-activating proteins (GAPs), which stimulate GTP hydrolysis and inactivation.

ARFmediated processes are widespread throughout eukaryotic cells. For instance, ARF1, a well-characterized member of the ARF family, plays a key role in the formation of coat proteins like COPI and clathrin, which are essential for the budding and trafficking of vesicles from the Golgi apparatus and endosomes. Other ARFs are involved in regulating the assembly of actin filaments, which are critical for cell shape, motility, and intracellular transport. Dysregulation of ARFmediated pathways has been implicated in various diseases, including cancer and viral infections, highlighting their importance in cellular homeostasis.
