Tropomyosin is a protein found in muscle and other cell types. In muscle cells, tropomyosin is a component of the thin filaments of sarcomeres, along with actin and troponin. It is a coiled-coil dimer that runs along the groove of the actin filament. In its relaxed state, tropomyosin physically blocks the binding sites on actin for the myosin heads, preventing muscle contraction. During muscle contraction, calcium ions bind to troponin, causing a conformational change that moves tropomyosin away from the actin binding sites. This allows myosin to bind to actin and initiate the cross-bridge cycle, leading to muscle shortening.
