Subunitpreferring is the property of a molecule that exhibits preferential binding or activity toward a specific subunit within a multimeric protein complex. The term is used across biochemistry, pharmacology, and structural biology to describe subunit specificity or selectivity. In receptor pharmacology, many receptors are heteromeric, composed of multiple subunit types; ligands may show higher affinity for receptor subtypes defined by subunit composition. This subunit bias influences pharmacodynamics, tissue distribution, and therapeutic profile.

Examples include benzodiazepines that preferentially modulate GABA_A receptors containing the alpha1 subunit (for example zolpidem), versus other alpha subunits. In the glutamate receptor family, ifenprodil preferentially antagonizes NMDA receptors containing the NR2B subunit. Ion channels, transporters, and proteasome subunits also show subunit-specific interactions, including antibodies or peptides designed to recognize a particular subunit.

Mechanisms arise from structural differences at subunit interfaces creating distinct binding pockets or allosteric sites; subunit composition can alter receptor conformation, gating, or signaling coupling.
