Selectins are a family of cell adhesion molecules that play a crucial role in the immune response and inflammation. They are characterized by their ability to bind to carbohydrate ligands, specifically sialylated Lewis X (sLeX) and sialylated Lewis A (sLeA), which are expressed on the surface of leukocytes and other cells. Selectins are divided into three main subtypes: L-selectin, E-selectin, and P-selectin, each with distinct functions and tissue distributions.

E-selectin, also known as endothelial leukocyte adhesion molecule-1 (ELAM-1), is primarily expressed on the surface of endothelial cells in response to inflammatory stimuli. It is rapidly induced by cytokines such as tumor necrosis factor-alpha (TNF-alpha) and interleukin-1 beta (IL-1 beta), and its expression is transient, lasting only a few hours. E-selectin plays a critical role in the early stages of inflammation by facilitating the adhesion of leukocytes to the endothelium, a process known as leukocyte rolling. This interaction is essential for the subsequent transmigration of leukocytes into the tissue, where they can participate in immune responses and tissue repair.

The binding of E-selectin to its ligands, sLeX and sLeA, is mediated by a lectin domain located in its extracellular region. This domain recognizes the specific carbohydrate structures on the surface of leukocytes, enabling the formation of stable adhesion complexes. E-selectin also interacts with other adhesion molecules, such as integrins, to enhance leukocyte-endothelial cell interactions.

The expression and function of E-selectin are tightly regulated, ensuring that its actions are confined to sites of inflammation and do not contribute to chronic inflammation or tissue damage. Dysregulation of E-selectin has been implicated in various pathological conditions, including atherosclerosis, autoimmune diseases, and cancer. Understanding the molecular mechanisms underlying E-selectin function is therefore of significant interest in the field of immunology and inflammation research.
