Proton transporter refers to any protein or protein complex that moves protons (H+) across biological membranes. By relocating protons, these transporters create or utilize a proton motive force (PMF) that drives many cellular processes, including ATP synthesis, nutrient uptake, and pH regulation.

Two broad categories exist. Primary proton pumps use energy from ATP hydrolysis or light to actively pump protons across membranes. Secondary proton-linked transporters rely on the existing proton gradient to move substrates in or out of the cell. Examples of primary pumps include F-type and V-type ATP synthases that can operate in reverse to pump protons, H+-ATPases in plants and fungi, and light-driven proton pumps such as bacteriorhodopsin found in certain archaea. Proton-linked transporters include symporters and antiporters that couple solute movement to proton translocation, enabling uptake of nutrients like sugars and amino acids in bacteria.

Mechanistically, most proton transport involves pathways formed by transmembrane helices with key acidic or basic residues that accept or donate protons as they move along. Proton movement can occur through direct translocation via a dedicated channel, or through transient conformational changes that shuttle protons across the lipid bilayer. The proton motive force comprises a chemical gradient (pH difference) and an electrical potential (membrane potential).

Physiological roles include energy transduction in respiration and photosynthesis, where PMF drives ATP synthesis in mitochondria and chloroplasts. Protons also power motility in certain bacteria and participate in nutrient uptake and pH homeostasis. Research focus includes understanding PMF generation, engineering proton-coupled transport systems, and developing optogenetic or biotechnological applications based on proton-transporting proteins.
