Proplastids are undifferentiated plastids that serve as precursors to the various plastid types found in plants. They are predominantly located in meristematic tissues, such as the shoot and root apical meristems, and in developing seeds. In plant cells they represent an early stage of plastid biogenesis before light-driven differentiation occurs.

Morphology and composition: Proplastids are small, typically lens-shaped organelles enclosed by a double membrane. They contain limited internal membrane organization and a modest amount of plastid DNA, ribosomes, and import machinery, reflecting their undifferentiated state. Their structure and activity are influenced by the cellular environment and exposure to light.

Differentiation pathways: In light, proplastids differentiate into chloroplasts with thylakoid membranes and grana, enabling photosynthesis. In dark conditions, they can become etioplasts, characterized by a prolamellar body and restricted photosynthetic capacity. In non-photosynthetic tissues, proplastids may differentiate into leucoplasts, which store fats, proteins, or starch, or chromoplasts, which accumulate pigments such as carotenoids. The transition between states is coordinated by plastid- and nucleus-encoded genes and involves extensive protein import and membrane development.

Role and significance: Proplastids are central to plastid diversification and plant development. They participate in the biosynthesis of fatty acids, amino acids, nucleotides, and pigments, and they can provide starch storage upon maturation. Retrograde signaling from plastids to the nucleus helps regulate nuclear gene expression in response to plastid status. In research and biotechnology, understanding proplastid development informs tissue culture practices and plastid transformation strategies.
