Prointerleukin-1beta refers to the inactive precursor form of interleukin-1beta (IL-1β), a critical cytokine involved in inflammatory responses. In its pro form, IL-1β is a 31 kDa protein that resides in the cytoplasm of cells, primarily monocytes and macrophages. It is synthesized from a messenger RNA transcript and accumulates within the cell without being secreted or exhibiting biological activity.

The activation and subsequent maturation of pro-IL-1β into its biologically active 17 kDa form is a tightly regulated process. This conversion is primarily mediated by caspase-1, an inflammatory protease.
