Procoagulants are substances that promote or accelerate blood coagulation, the process by which blood changes from a liquid to a gel, forming a blood clot. This physiological process is essential for hemostasis, preventing excessive bleeding after injury. Procoagulants can be naturally occurring components of the blood clotting cascade or externally administered agents.

Within the body, the coagulation cascade is a complex series of enzymatic reactions involving numerous clotting factors. Many of these factors, when activated, act as procoagulants by initiating or amplifying the cascade. For example, thrombin is a key procoagulant enzyme that converts fibrinogen into fibrin, the protein meshwork that forms the structural basis of a blood clot. Other intrinsic and extrinsic pathway factors also play crucial procoagulant roles.

Medically, procoagulant agents are used to treat bleeding disorders or excessive blood loss. These can include specific clotting factor concentrates, such as those for hemophilia, or medications that stimulate the production of clotting factors. Some topical agents applied to wounds can also function as procoagulants, aiding in rapid clot formation. The use of procoagulants is carefully managed to avoid unwanted thrombosis, the formation of clots within intact blood vessels.
