Plasminmediated refers to biological processes driven by plasmin, a broad-spectrum serine protease central to fibrinolysis and extracellular matrix remodeling. Plasmin is generated from its zymogen plasminogen by activators such as tissue-type plasminogen activator (tPA) and urokinase-type plasminogen activator (uPA).

In the circulatory system, plasmin enzymatically degrades fibrin clots, facilitating thrombus resolution. Beyond fibrinolysis, plasmin degrades several extracellular matrix components (including fibrin, fibronectin, laminin, and collagen IV) and can activate matrix metalloproteases, contributing to tissue remodeling, wound healing, angiogenesis, and cell migration. Plasmin can also release and activate growth factors and modulate inflammatory processes by processing cytokines and complement components.

Activity is tightly regulated by inhibitors such as alpha-2-antiplasmin, alpha-2-macroglobulin, and plasminogen activator inhibitors (PAI-1, PAI-2), as well as by C1-inhibitor and α2-antiplasmin. The balance between plasmin generation and inhibition determines fibrinolytic potential and proteolytic remodeling in tissues.

Clinically, plasmin-mediated pathways are targeted in thrombolytic therapy using plasminogen activators (e.g., tPA) to dissolve clots in acute ischemic stroke, myocardial infarction, and pulmonary embolism. Dysregulation of plasmin activity is implicated in bleeding disorders, thrombosis, cancer invasion, fibrosis, and inflammatory diseases. Experimental research continues to define plasmin’s diverse substrates and contextual roles in physiology and pathology.
