Plant protoplasts are plant cells from which the cell wall has been removed. This process is typically achieved enzymatically using cell wall-degrading enzymes like cellulase and pectinase. The resulting structures are osmotically sensitive and must be maintained in an isotonic solution to prevent bursting. Protoplasts are valuable tools in plant biology and biotechnology due to the absence of their rigid cell wall, which normally restricts cellular processes.

The isolation of protoplasts opens up a range of applications. They are crucial for plant somatic hybridization, where protoplasts from different plant species are fused to create hybrid cells that can potentially regenerate into novel plant varieties. This technique bypasses the sexual incompatibility that often prevents hybridization between distant species. Protoplasts are also widely used in genetic transformation. Their naked state allows for the direct introduction of foreign genetic material, such as DNA or RNA, through methods like electroporation, polyethylene glycol (PEG) treatment, or microinjection.

Furthermore, protoplasts serve as excellent models for studying various cellular processes, including membrane transport, signal transduction, and gene expression, without the interference of the cell wall. They can be cultured and regenerated into whole plants under specific laboratory conditions, enabling the study of plant development and the generation of genetically modified plants.
